Synaptic and network mechanisms of motor timing

Flexible control of movement onset, or motor timing, is crucial for virtually all behaviors. Before voluntary movements begin, both the frontal cortex and striatum exhibit ramping spiking activity, where the slope of the ramp varies across trials and anticipates movement onset, functioning as an adjustable 'timer' for motor timing. However, the computational roles mediated by each area to implement this timer function, whether distributed, redundant, or specialized, remain unclear due to their similar dynamics and indispensability. Additionally, how such dynamics emerge during learning is largely unknown. I will present two projects from my new lab that address these critical unknowns. We leveraged molecular manipulation of synaptic plasticity, transient/calibrated optogenetic perturbations, and large-scale electrophysiology in mice learning and performing a flexible lick timing task. Our findings 1) support a model where the striatum serves as a timer via integrator dynamics, generating ramping activity that governs motor timing, while the frontal cortex provides input to adjust the timer, and 2) identify a key neocortical cell type in which synaptic plasticity shapes cortical dynamics to learn new lick timing.
